Selank — Anxiolytic Heptapeptide

Background

Selank is a synthetic heptapeptide (Thr-Lys-Pro-Arg-Pro-Gly-Pro) developed at the Russian Institute of Molecular Genetics in collaboration with the V.V. Zakusov Research Institute of Pharmacology. It is structurally modelled on tuftsin (Thr-Lys-Pro-Arg), a naturally occurring immunomodulatory tetrapeptide.

Mechanism

Published Russian work attributes Selank's effects to enkephalinase inhibition (stabilising endogenous enkephalins), modulation of GABAergic and serotonergic neurotransmission, and BDNF upregulation in the hippocampus.

What is proven (largely in Russian literature)

Russian preclinical and limited clinical data report anxiolytic activity comparable to benzodiazepines (medazepam) in generalised anxiety disorder, without sedation, cognitive impairment, withdrawal symptoms or dependence liability. Cognitive performance and mnemonic function improvements have been reported.

Intranasal administration is the standard route in published Russian work, with reasonable nose-to-brain delivery.

What is NOT proven

Western RCTs are essentially absent. Long-term human safety data outside Russian post-marketing experience are not characterised. Cognitive enhancement in healthy individuals is not robustly demonstrated by Western standards.

Pharmacokinetics in non-intranasal routes; reproducibility of Russian clinical data in Western trial conditions; long-term BDNF effects.

Regulatory status

Registered in Russia for anxiety disorders. Not approved by the FDA, MHRA, or EMA.
