    GHK-Cu — Copper Tripeptide-1

    Background

    GHK-Cu is a complex of the tripeptide glycyl-L-histidyl-L-lysine (GHK) with a copper(II) ion. GHK was originally isolated from human plasma by Loren Pickart in 1973. Plasma levels are approximately 200 ng/mL in healthy 20-year-olds and decline by roughly 60% by age 60.

    Mechanism

    The GHK sequence binds copper with very high affinity, and the resulting GHK-Cu complex serves as a copper-delivery molecule to skin and other tissues. Documented downstream effects include stimulation of dermal fibroblast proliferation, collagen and elastin synthesis, glycosaminoglycan and decorin production, antioxidant gene expression (SOD, catalase), and modulation of metalloproteinase activity.

    A widely-cited 2010 genomic analysis (Pickart, Vasquez-Soltero, Margolina; published in Biomed Research International / OA literature) reported that GHK modulated expression of more than 4,000 human genes toward a younger expression profile.

    What is proven

    Topical GHK-Cu has demonstrated improvements in skin elasticity, fine wrinkles, photodamage and wound healing in multiple controlled cosmetic dermatology trials. It is a long-standing active ingredient in licensed cosmetic formulations worldwide.

    Injected GHK-Cu has shown accelerated wound closure, hair-follicle stimulation, and antioxidant activity in animal models. Mechanistic effects on fibroblast and stem-cell signalling are well-replicated.

    What is NOT proven

    Robust human trials for systemic 'anti-ageing' endpoints (lifespan, biological age clocks, organ function) following injected GHK-Cu are not available. Hair-regrowth claims for injected GHK-Cu rest on small-scale studies and extrapolation from topical/scalp formulation data.

    Optimal injected dose, frequency and copper-loading thresholds in humans are not characterised. Copper accumulation risk with chronic high-dose use has not been formally studied.

    Open research questions

    Systemic versus local effects of injected GHK-Cu, the safety ceiling of repeated copper-bound peptide dosing, and whether genomic-modulation findings translate to durable phenotypic change in humans all remain open.

    Regulatory status

    Approved as a cosmetic ingredient in most jurisdictions. Not approved by any major regulator as an injectable therapeutic for any indication.
