Background
This combination pairs CJC-1295 No DAC (a GHRH analogue) with Ipamorelin (a selective ghrelin receptor agonist). It is the standard reference combination cited in GH-secretagogue research because the two compounds act on different pituitary receptors with complementary mechanisms.
Mechanism
GHRH analogues amplify pituitary somatotroph readiness to release GH; ghrelin receptor agonists trigger the release. Combined administration produces a greater GH pulse than either compound alone — a well-documented pharmacological synergy.
What is proven
Synergy between GHRH and GHRP class compounds at the pituitary is established in human pharmacology studies. Combined GH pulse amplitude exceeds additive expectation. IGF-1 elevation is dose-dependent.
What is NOT proven
No completed Phase 3 outcome trials of this specific combination. Long-term human safety, body-composition and healthspan endpoints rest on pharmacology, not on registrational data.
Open research questions
Whether amplified pulsatile GH release confers durable body-composition or recovery benefits beyond what selective single-receptor agonism delivers; long-term IGF-1 elevation surveillance.
