    5-Amino-1MQ — The NNMT Inhibitor

    Background

    5-Amino-1-methylquinolinium (5-Amino-1MQ) is a small-molecule selective inhibitor of nicotinamide N-methyltransferase (NNMT), an enzyme that catalyses the methylation of nicotinamide using S-adenosylmethionine (SAM). NNMT is upregulated in white adipose tissue in obesity and in certain cancers.

    Note: 5-Amino-1MQ is not a peptide — it is a quinolinium small molecule. It is grouped with metabolic research compounds because its target lies in obesity and metabolic biology.

    Mechanism

    NNMT inhibition by 5-Amino-1MQ raises intracellular nicotinamide and SAM levels, increases NAD+ availability, suppresses adipocyte differentiation, and stimulates lipolysis in white adipose tissue. Effects on muscle mitochondrial function have also been reported.

    What is proven

    Multiple rodent studies have demonstrated reduced fat-mass gain on high-fat diet, prevention of diet-induced obesity, preservation of lean mass, and improvements in insulin sensitivity and metabolic markers without changes in food intake. Mechanistic effects in cultured adipocytes are well-replicated.

    What is NOT proven

    No completed human efficacy or safety trials. Whether the rodent fat-loss effect translates to humans at safe doses is unstudied. Long-term safety, oncology signal (NNMT is upregulated in several cancers, so inhibition is plausibly beneficial but unstudied chronically), and optimal dosing are unknown.

    Open research questions

    Tissue selectivity of NNMT inhibition; whether peripheral NNMT inhibition is sufficient or central activity is required; chronic safety profile; interaction with weight-loss agents acting on appetite (GLP-1 class).
